Hydrolyzed Casein and Sleep Quality: Evidence from Animal Studies to Human Research

2026-02-24

With the fast pace of modern life, sleep disorders have become a widespread health problem. Naturally derived bioactive ingredients are increasingly becoming an important research direction in sleep management. Among them, hydrolyzed casein and its derivatives have received increasing attention due to their good safety profile and multi-pathway regulatory capabilities.

Why hydrolyzed casein?

Casein is the main protein source in dairy products. After targeted enzymatic hydrolysis, it can release a variety of bioactive peptides with neuromodulatory potential. Unlike intact proteins, the small peptides in hydrolyzed casein are more easily absorbed and may affect sleep-related physiological processes through the gut-brain axis, neurotransmitter regulation, and endocrine pathways.Compared to single-target components such as melatonin, the effects of hydrolyzed casein are more systemic, which is a key reason for its continued popularity.

Animal Experiments: Regulation of Sleep Structure by Hydrolyzed Casein

1. Improvement of Slow-Wave Sleep (SWS) Quality

Multiple animal studies have found that supplementation with casein hydrolysates of a specific degree of hydrolysis can significantly affect the sleep structure of rats, particularly in the following ways:

Prolonging the duration of slow-wave sleep (SWS)

Improving sleep continuity and stability

Reducing the number of nighttime awakenings

Slow-wave sleep is a phase in the sleep cycle closely related to bodily repair, immune regulation, and memory consolidation. Its decreased quality is often associated with chronic fatigue and weakened immune function.

2. Improvement of Abnormal Sleep

In stress or sleep rhythm disruption models, hydrolyzed casein has been found to:

Improve abnormal REM sleep duration in rats

Alleviate experimentally induced sleep disturbances

These results suggest that hydrolyzed casein may have a positive impact on overall sleep by regulating the excitation-inhibition balance of the central nervous system.

Evidence from Human Studies

1. Significant Improvement in Sleep Quality Index (PSQI)

A human intervention study conducted in Japan showed that after continuous supplementation with hydrolyzed casein, the total score of the Pittsburgh Sleep Quality Index (PSQI) in subjects significantly decreased, indicating an improvement in overall sleep quality.

After 2 weeks of treatment: Overall sleep quality began to improve.

After 4 weeks of treatment: Sleep latency shortened (easier to fall asleep).

Daytime dysfunction significantly reduced.

Sleep satisfaction improved.

Furthermore, the study did not report any significant adverse reactions, demonstrating good safety and tolerability.

2. Advantages

Unlike traditional sedative sleep aids, hydrolyzed casein did not cause significant drowsiness, dizziness, or daytime cognitive impairment. This makes it more suitable for long-term use in functional foods and daily nutritional supplements.

How Does Hydrolyzed Casein Affect Sleep?

Current research suggests that its mechanism of action may include:

1. Neurotransmitter Regulation

Some peptides released from hydrolyzed casein may affect the GABAergic nervous system, helping to reduce neural excitability and promote relaxation.

2. Gut-Brain Axis Involvement

Small molecule peptides may indirectly participate in sleep rhythm regulation in the gut by influencing gut microbiota metabolism or neuroendocrine signaling.

3. Anti-inflammatory and Antioxidant Support

Chronic inflammation and oxidative stress are considered important triggers for sleep disorders. Hydrolyzed casein has been shown to possess certain anti-inflammatory and antioxidant activities, potentially providing indirect support for improving sleep.

Application Prospects

Based on current research results, hydrolyzed casein has the following promising application potential:

Sleep-supporting functional foods

Stress management and recovery nutritional supplements

Nutritional formulas for middle-aged and elderly individuals

As a mild and long-term usable base ingredient

Increasing research evidence suggests that hydrolyzed casein is not only a high-quality protein source but also a functional ingredient with sleep regulation potential. Its specific mechanisms are still under investigation, but its safety and multi-pathway regulatory characteristics give it long-term development potential.
