What is Glucanase?
Glucanase(Beta Glucanase) belongs to the class of hydrolases. Its main function is to catalyze the hydrolysis of glycosidic bonds in β-glucan molecules. Its core function lies in effectively breaking down the high-viscosity β-glucan in grains such as barley and oats, thereby significantly reducing the viscosity of gels formed during processing or digestion. This characteristic makes it valuable and widely used in food (such as beer brewing), feed, and pharmaceutical industries. Furthermore, this enzyme also shows broad application potential in sugar refining, textiles, papermaking, and even oral care (such as breaking down dental plaque).

Mechanism of Action
It can be summarized in the following two key steps:
Endolysis
The glucanase molecule recognizes and adsorbs onto long-chain β-glucan through its substrate-binding domain. Subsequently, the active site in its catalytic domain (usually containing key amino acid residues such as glutamic acid and aspartic acid) randomly cleaves the β-1,3 or β-1,4 glycosidic bonds within the polysaccharide chain. This step rapidly cleaves the long-chain, high-viscosity β-glucan macromolecules into a series of short-chain oligoglucans, directly disrupting the structural basis for forming high-viscosity gels.
Exolytic Hydrolysis
After endolytic hydrolysis produces short-chain products, another type of exolytic glucanase can further act on the ends of these short chains, cleaving glucose units one by one. Ultimately, β-glucan is completely broken down into glucose that can be directly absorbed and utilized, as well as oligosaccharides with prebiotic functions.
Applications of Glucanase in Multiple Fields
Beer Brewing: This is the most important application of glucanase. During mashing, adding glucanase can degrade β-glucan in malt, significantly reducing wort viscosity, solving filtration difficulties, increasing filtration speed and wort yield, and improving the non-biological stability of beer.
Feed Additives: Glucanase is used as an important feed additive in feeds made from barley, wheat, etc. It can degrade β-glucan, which animals cannot digest, allowing digestive enzymes to fully contact nutrients, thereby improving the digestibility and absorption of energy and protein, which is beneficial to animal growth performance.
Sugar Production: When sugarcane juice viscosity abnormally increases due to microbial infection and the production of glucan, adding glucanase can effectively reduce viscosity and improve the juice's flowability and heating efficiency, thus shortening clarification and crystallization time and increasing production efficiency.
Food Processing: Glucanase is used for juice clarification and works synergistically with pectinase to increase juice yield.
Chemicals and Pharmaceuticals: Glucanase can be added to toothpaste to break down the glucan structure in dental plaque, aiding in oral hygiene; glucanase can also be used in drug sustained-release systems.
